Molecular and pharmacological study of transcription factor Nrf2: A master regulator of cellular antioxidant defense enzymes. $69 Nuclear factor E2-related factor 2 (Nrf2) is a key regulator of ARE-mediated gene expression of Phase II detoxifying enzymes and antioxidant enzymes. In the proposed study, we tested whether nuclear receptor-associated coactivator-3 (RAC3) could participate in the stimulation of transactivation activity of Gal4-Nrf 2 (1-370) with other coactivators. We showed for the first time that the p160 coactivator RAC3 could stimulate the transactivation activity of Gal4-Nrf 2 (1-370) and this could be further enhanced by the coregulators CBP, p/CAF, CARM-1, and PRMT1. Besides, we found that regulation of Nrf2-dependent ARE-driven gene expression by small Mafs protein is dependent on dosage and the ratio of Maf to Nrf2. On the other hand, ERK2 significantly enhanced the expression of ARE-driven luciferase. ERK2 and MafG synergistically upregulated the Nrf2-activated ARE-driven luciferase expression. Nrf2 was identified as a direct substrate of ERK2 both in vivo as well as in vitro for the first time. The phosphorylation of Nrf2 by ERK2 increased its heterodimerization with small Maf proteins. To extrapolate the implication of Nrf2 in inflammation, we compared the anti-inflammatory effect of sulforaphane on LPS-stimulated inflammation in primary peritoneal macrophages derived from Nrf2 (+/+) and Nrf2 mice. The anti-inflammatory effect was attenuated in Nrf2 (-/-) primary peritoneal macrophages. We concluded that sulforaphane exerts its anti-inflammatory activity mainly via activation of Nrf2 in mouse peritoneal macrophages. To better facilitate application of sulforaphane in therapeutics, the pharmacokinetics characterization was investigated in rats. Prediction Method Of Air Pressure On 2-Pipe Stack Building Drainage $64 This research first examines the current condition of the building drainage system design in Taiwan, including an investigation 51 apartment buildings. The second part is to experiment on the air pressure in the vertical stack drainage system . An experimental device that simulates a medium-height apartment was set up to provide empirical parameters and model verification at NTUST. A considerable progress has been made in predicting the air pressure distribution within vertical drainage stacks.The third part focuses on a 2-pipe vertical drainage stack for conducting the empirical parameter and evaluation method of air pressure of building drainage systems. Next, the air pressure prediction in 2-pipe vertical drainage stacks from the predicted results of a single drainage stack has been illustrated.The study also conducts the comparison between the measured data and calculated values. It reveals that these prediction and evaluation methods can reproduce the mean air pressure distribution value in a vertical drainage stack. Rainbow: Cost-Effective software architecture-based self-adaptation. $69 Modern, complex software systems (e-commerce, IT, critical infrastructures, etc.) are increasingly required to continue operation in the face of change, to self-adapt to accommodate shifting user priorities, resource variability, changing environments, and component failures. While manual oversight benefits from global problem contexts and flexible policies, human operators are costly and prone to error. Low-level, embedded mechanisms (exceptions, time-outs, etc.) are effective and timely for error recovery, but are local in scope to the point-of-failure, application-specific, and costly to modify when adaptation objectives change. An ideal solution leverages domain expertise, provides an end-to-end system perspective, adapts the system in a timely manner, and can be engineered cost-effectively. Architecture-based self-adaptation closes the "loop of control," using external mechanisms and the architecture model of the target system to adapt the system. An architecture model exposes important system properties and constraints, provides end-to-end problem contexts, and allows principled and automated adaptations. Existing architecture-based approaches specialize support for particular classes of systems and fixed sets of quality-of-service concerns; they are costly to develop for new systems and to evolve for new qualities. To overcome these limitations, we posit this thesis: We can provide software engineers the ability to add and evolve self-adaptation capabilities cost-effectively, for a wide range of software systems, and for multiple objectives, by defining a self-adaptation framework that factors out common adaptation mechanisms and provides explicit customization points to tailor self-adaptation capabilities for particular classes of systems, for multiple quality-of-service objectives.
